Work by PhD student Lara Macheriotou and colleagues resulted in the first publication of the year 2019 for the Marine Biology Research Group. The article describes a methodological investigation of the utility of our internal curated nematode reference sequence database for the definition of species-delimiting Operational Taxonomic Unit (OTU) clustering thresholds by quantifying intra- and interspecific genetic differentiation with p-distance. The derived thresholds were subsequently tested on artificial communities as well as the performance of the clustering algorithms Uparse, Uclust_ref, the cluster-free Dada2 algorithm and the RDP taxonomic classifier given differing reference databases.
